International Microarray Workshop: May 2005


 

PLANT LONG-OLIGONUCLEOTIDE
MICROARRAY WORKSHOP

This workshop comprised a combination of lectures and hand-on laboratory sessions.  The participants primarily employed Arabidopsis and maize whole genome 70-mer oligonucleotide microarrays in their laboratory work (for details of these arrays see http://www.ag.arizona.edu/microarray and http://www.maizearray.org/).  The workshop was aimed at the goal of obtaining optimal results using oligonucleotide-based microarrays.  It was offered concurrently with a workshop focusing on animal long oligonucleotide microarrays.

The workshop was divided into two parts:  Part I covered wet-lab aspects of microarray target production, microarray hybridization, and scanning.  It started with registration on Sunday, May 8, from 2-4PM, with the first formal activities at 5PM, and ended on Wednesday evening.  Part II covered data extraction, statistical analysis, and experimental design, and occupied Thursday and Friday.  Part I activities (plant and animal systems) were offered in separate laboratories, whereas Part II activities were offered jointly.

Specific topics covered included:

  • Experimental design.
  • Probe preparation and microarray printing.
  • Microarray rehydration and probe immobilization.
  • Target preparation, including RNA extraction, direct and indirect labeling, and amplification techniques.
  • Microarray hybridization.
  • Array scanning and data extraction.
  • Data analysis and archiving.
